Enfrente de la Plaza de Santo Domingo se puede apreciar otra de las bellezas arquitectónicas que hay en el Centro Histórico. La Casa del Mayorazgo de Medina cuenta con intervenciones del barroco y el siglo XVIII. Ahí vivió Diego de Pedraza en 1525, el primer cirujano de la Nueva España. Posteriormente perteneció al Mayorazgo de Medina, de ahí el nombre. Actualmente hay comercios en su interior.
